چکیده

It has been clearly established (1, 2), through the use of N16-labeled material, that the utilization of free thymine for the synthesis of deoxyribonucleic acid (DNA) by the rat is extremely limited. However, these studies afforded but little information concerning the rate and extent of the metabolism of thymine. In the experiments reported here, thymine-204 has been found to be more rapidly and extensively catabolized by this species than was heretofore appreciated. In marked contrast to the results obtained after administration of uracil-N15 and thymine-N15 to rats (2), and of uracil-N15 to human subjects (3), only a small amount, of the tracer was found to be excreted via the urine. A large portion (55 per cent) of the isotope was excreted, however, in the respiratory COz during a period of 4 hours following intraperitoneal administration of thymine-2-Cl4. Experiments with rat liver slices also have shown that thymine-2-C*4 is rapidly catabolized by this tissue to give rise to C402. In agreement with the findings of Brown, Roll, and Weinfeld (I), a small but significant amount of the administered isotope was found in the thymine moiety of DNA of regenerating rat liver, and a lesser amount in the DNA of other tissues.

Certain interrelationships in the metabolism of glycine and serine.

The utilization of L-serine-2,3-deuterio, 3-C4, N16 and glycine-2-C4, N15 for the biosynthesis of purines, thymine, and choline in the rat was described briefly in preliminary publications from this laboratory (1, 2). In the synthesis of the methyl groups of choline from the p-C14DaOH group of isotopically labeled serine it was later found (3) that t,he C14:D ratio was unaltered, indicating tha...

The effect of chronic psychological stress on carbohydrate metabolism in rat

Several studies have shown that stress has major effects on metabolism. There is also some evidence indicating that stress may induce diabetes mellitus. The present study has investigated the role of chronic psychological stress on carbohydrate metabolism in male Wistar rats (250 ± 50 g). Animals were divided into control and test groups (n = 8/group). Blood samples were obtained by tail snippi...

Studies on fluorinated pyrimidines. V. Effects on nucleic acid metabolism in vitro.

To gain further information on the mechanisms by which the fluorinated pyrimidines and their nucleosides inhibit various processes of nucleic acid biosynthesis, studies similar to those re ported in the preceding paper (6) were carried out in vitro in glycolyzing suspensions of the Ehrlich ascites cells system used by LePage for studies of purine biosynthesis (20). In this work the following la...

Changes in Biochemical Parameters Related to Lipid Metabolism Following Lithium Treatment in Rat

Lithium is widely used in medicine as an anti-depressive drug. In spite of abundant literature, questions on the side effects of lithium ions are far from being answered. In this study, the effects of lithium on biochemical parameters related to lipid metabolism were investigated. Male Wistar rats were treated with different doses of lithium for a period of up to 60 days. Blood samples were col...

Effect of 5-bromodeoxyuridine on deoxyribonucleic acid-thymine synthesis and cell metabolism of lymphatic tissues and tumors.

5-Bromodeoxyuridine (BrUDr) is an effective inhibitor of the growth of E. coli and Lactobacitti, and this growth inhibition can be reversed by thymidine (1-3). There is evidence that this thymidine analog can replace the thymine of both E. coli and bacteriophage deoxyribonucleic acid (DNA) on an equimolar basis (7, 11, 12). In E. coli, nearly 50 per cent of the thymine can be replaced. Whereas ...
